Simona Suchá, Ph.D. Title of diploma thesis: Characterization of gilteritinib-resistant leukemic cell line Acute myeloid leukemia (AML) is an aggressive cancer with a poor prognosis for which therapy failure is often responsible. Development of drug resistance is among the most common causes of treatment failure. The exact mechanisms leading to the resistance are not known, especially for drugs recently introduced into the clinical practice such as gilteritinib. Therefore, the gilteritinib-resistant leukemic cell line (referred to as HL-60 g75) has been established in our lab and further characterized. The aim of this study was to evaluate the sensitivity of HL-60 g75 cells to gilteritinib and to clarify the stability of acquired resistance. We revealed that the resistance of HL-60 g75 cells is transient, since only after 4 weeks of gilteritinib-free cell culture, sensitivity of these cells was restored. While gilteritinib induced apoptosis similarly in both gilteritinib-sensitive (HL-60 wt) and -resistant cells, cell cycle analysis revealed lower responsiveness of HL-60 g75 to gilteritinib than HL-60 wt....

Unfortunately the serious and life-threatening adverse effect cardiotoxicity compromises their clinical usefulness. The only approved protection against anthracycline cardiotoxicity so far is dexrazoxane. Despite the outstanding cardioprotective ability, dexrazoxane use is very limited mainly due to its possible side effects. So we were directed towards synthesis of dexrazoxane analogues with better pharmacological properties. Genetic information must be protected, maintained and copied from cell to daughter cells, from generation to generation. In plants, most of the cells contain complete genetic information, and many of these cells can regenerate to a whole new plant. Such a feature leads to the need for precise control of which genes will be active and which not because in growth and differentiation, only the activity of specific genes for the individual cells, tissues, organs are required. One of the mechanisms controlling the gene activity is RNA interference (RNAi), which down- regulates or blocks the expression of specific genes at the transcriptional or post-transcriptional level. The crucial part of the RNAi is guiding the RNAi machinery to the target. It is mediated via sequence complementarity of the target with a small RNA (sRNA), which is diced from a double- stranded RNA (dsRNA) precursor. The molecular mechanism of dsRNA and sRNA formation and also the target origin predestinates the subsequent silencing pathway. In transcriptional gene silencing (TGS), the gene expression is regulated through chromatin epigenetic modifications. Návrh a testování způsobů zvýšení účinnosti transfekce živočišných buněk

Master’s thesis deals with the design and testing of methods of increasing the transfection efficiency in mammalian cells. In this work a polyethylenimine (PEI) transfection method was used. The transfection efficiency was tested using MDA-MB-231 breast cancer cell line and pcDNA3.1-GFP-hMT-TOPO vector. We focused on optimization of PEI (1-6 µg) and plasmid DNA (1-3 µg) concentration, transfection time (0-48 h), number of cells per 9 cm2 (100,000-2 million) and medium composition. A complex containing linear penetration peptides (CPPs) KALA and GALA was assembled. Concentration of CPPs (7,5-45 µg) was optimized and a zeta potential and a size of the complexes were investigated. Due to the formation of large aggregates (over 1200 nm), modification with polyethyleneglycol (PEGylation) of PEI was performed. With the resulting complexes (~15 nm), transfection was performed. Transfection and apoptosis induction rates were compared between PEI and PEGylated PEI and as the best was chosen conjugate of PEGylated PEI (3 µg) with 45 µg of GALA and 3 µg pDNA with efficiency of 75%.

Establishment and characterization of novel IL-6-dependent myeloma cell line ÚHKT-893

Multiple myeloma is an incurable fatal neoplasm of plasma cells affecting mainly elderly people. There are many research laboratories in the world where multiple myeloma is studied. Permanent cell lines are indispensable tools for both basic and applied research. However, the establishment of new cell lines is difficult with poor success. We established 96 primary cultures of bone marrow samples from myeloma patients. Only one culture succeeded in permanet myeloma cell line. The novel plasmacytic cell line ÚHKT-893 was established from bone marrow sample of 57 years old female relapsed with multiple myeloma of IgGκ isotype. The cell line is however dependent on the continuous presence of interleukin-6 in culture media. ÚHKT-893 cells grow continuously already more than 1 year. The growth of cells was regularly monitored according to growth curves and by measurement of cell viability. Surface antigenic profile was repeatedly determined by flow cytometry. The simultaneous expression of both CD138 and CD38 surface molecules confirmed the plasma cell origin of cells. The establishment of the ÚHKT-893 myeloma cell line will extend the panel of existing myeloma cell lines as a new ex vivo model for the study of the etiology and pathogenesis of multiple myeloma. The effect of IDGF2 protein on expression of selected genes in cell line Cl.8+

The imaginal discs growth factors (IDGFs) are proteins able to induce proliferation of Cl.8+ cells proliferation in collaboration with insulin. They are also connected with response to starvation stress and innate immunity. A genome-wide analysis of Cl.8+ cells treated with IDGF2, which was performed in our lab, detected tens of genes transcriptionally induced by this protein. The aim of this work was to confirm this effect with some of these genes using quantitative PCR.
